Cells were exposed to two ATP release\nstimuli: 3V and the peptide mastoparán 7 (MST7). Both stimuli induce increases\nof cAMP, the main second messenger.\nMain results:\nUnder 3V exposure there is an increase of cAMP, which translated into an\nacute, transient release of ATP. When cells were pre-exposed to CBX\n(pannexin 1 inhibitor), ATP release induced by 3V was completely blocked.Thus\nunder 3V stimulation, RBCs exhibit a single ATP conduit mediated by pannexin\n1.\nUnder MST7 exposure, small cAMP increases correlated with a strong ATP\nrelease. CBX induced a partial blockage of ATP release, suggesting the\nexistence of two conduits for ATP release. In addition to cAMP, the potential\nrole of cell volume and cell adhesion on ATPe kinetics was studied.\nIn non-adhered cells, MST7 neither trigger ATP release nor affected cell\nvolume. In vitro, la\nexposición a diferentes agentes farmacológicos desencadena eventos intracelulares\nque activan la liberación de ATP.\nEn esta tesis se analizaron los factores que controlan la evolución temporal de la\nconcentración de ATP extracelular (ATPe). Las células fueron expuestas a dos\nestímulos que inducen liberación de ATP: 3V y mastoparán 7 (MST7). Ambos\nestímulos inducen aumentos de la concentración de cAMP.\nLa exposición a 3V produjo un aumento de cAMP, que se tradujo en una liberación\naguda de ATP. Cuando las células fueron tratadas con CBX (bloqueante de\npanexina 1), la liberación de ATP fue completamente bloqueada concluyendo que la\nsalida de ATP ocurriría sólo a través de la panexina 1.\nLa exposición a MST7 indujo pequeños aumentos de cAMP que llevaron a una gran\nsalida de ATP. El CBX bloqueó parcialmente la liberación de ATP, sugiriendo la\nexistencia de dos vías para la liberación del mismo. Además se estudió el papel del\nvolumen y la adhesión celular en la cinética de ATPe.
