As study models, tumor cells in culture (HeLa, B16-F0, B16-F10, MCF-7 and MDA-MB-231), arrested in G1/S phase of the cell cycle, were used. In cells treated with DhL were analyzed the properties of adhesion, invasion and migration. DhL did not induce apoptosis or senescence and neither affected the cell viability. DhL inhibited the matrigel invasion and the migration in a wound healing assay. DhL inhibited the membrane protrusion in the cell front due to affected the organization of actin cytoskeleton and inhibited the signalling proteins as Rac1, ERK and p38. These results indicated that DhL, affects the adhesion, invasion and migration of tumor cells in culture due to the inhibition of ERK signalling pathway. This work provides valuable information to consider DhL as a potential pharmacology drug to be used as a antimetastatic agent in cancer therapies. Fil: Amaya, Celina. Universidad de Buenos Aires. Previamente, nuestro laboratorio ha demostrado que DhL inhibe la proliferación de células tumorales en cultivo y el crecimiento de tumores en ratones. Con el fin de demostrar que DhL posee potencial para el desarrollo de fármacos antitumorales, en esta tesis analizamos las propiedades antimigratorias de DhL en células tumorales. Como modelo de estudio utilizamos células en cultivo, arrestadas en la fase G1/S del ciclo celular. En las células tratadas con DhL fueron analizadas las propiedades de adhesión, invasión y migración. Como resultado, observamos que DhL no indujo apoptosis ni senescencia y no afectó la viabilidad celular. Luego, en una superficie recubierta con fibronectina, comprobamos que DhL inhibió la adhesión y la formación de adhesiones focales. Posteriormente, observamos que DhL inhibió la invasión del matrigel por las células tumorales y la migración celular. Además, DhL inhibió la protrusión de la membrana en el frente de avance, por la alteración de la organización del citoesqueleto de actina y la actividad de proteínas señaladotas como Rac1, ERK y p38. Estos resultados indican que DhL, a través de la inhibición de la señalización de ERK, afecta la adhesión, invasión y migración de las células tumorales en cultivo, confirmando la hipótesis que esta lactona sesquiterpénica tiene un gran potencial farmacológico como agente antimetastásico.
