We used\nviral vectors that express anti-sense sequences against the ARNms of the GluN2A and\nGluN2B subunits. These vectors were inoculated into the hippocampus using estereotaxic\nsurgery in Wistar rats, and their effects in learning and memory were evaluated using Open\nField (OF) and Inhibitory Avoidance (IA) tests. The expression of the transgenes was\nevaluated by ?Western blot" and "Polimerase Chain Reaction?. We observed a significant\ndecrease of the expression of GluN2A, in the group of rates inoculated with vectors that\nexpressed sequences anti-sense against this subunit. Es uno de\nlos principales tipos de receptores que median la transmisión especial por glutamato, aunque\nno la neurotransmisión corriente, la cual se encuentra mediada por receptores de tipo AMPA.\nEl RNMDA se expresa en todo el sistema nervioso central, en especial en el hipocampo,\nestructura cerebral que participa en la formación de memorias espaciales dando lugar a\nmemorias de largo plazo.\nEn este trabajo se estudió el rol de las subunidades regulatorias (GluN2A y GluN2B) del\nRNMDA en el establecimiento de nuevas memorias, mediante la disminución de la expresión\nde las mismas. Para ello se utilizaron vectores virales que expresan secuencias anti-sentido\ncontra los ARNms de las subunidades GluN2A y GluN2B. Estos vectores fueron inoculados\nintrahipocampo mediante cirugía estereotáxica en ratas de la cepa Wistar y se evaluó su\nefecto sobre el aprendizaje y la memoria a través de los test comportamentales de Campo\nAbierto (CA) y Evitación Inhibitoria (EI). La expresión de los transgenes se evaluó mediante\ntécnicas de biología molecular, Western blot o Polimerase Chain Reaction. Se observó una\ndisminución significativa de la expresión de GluN2A, en el grupo de ratas inoculadas con\nvectores que expresaron secuencias anti-sentido contra esta subunidad.
