An analysis of Franciscan bishop Bernardino de Cárdenas and the Jesuits fathers for the control of the guaraní settlments during the 1640' decade is done. Also opposing political and economic interests which divided Asuncion society into two groups are untangled. In other words, a general social conflict arose in defense of maintaining privileges already granted. This controversial issue with strong political repercussions allows us to visualize the problem of indigenous labor and the jesuit-guaraní alliance. This in turn lead us to understand the impact of the spiritual conquest in the new social order.  El presente trabajo tiene por objeto investigar en la región del Paraguay el impacto de la creación del sistema de reducciones jesuíticas sobre las relaciones interétnicas hispano-guaraníes durante la priera mitad del siglo XVII. A tal efecto se analiza el conflicto religioso que enfrentó durante la década de 1640 al obispo franciscano Bernardino de Cárdenas y a los padres de la Compañía por el contro de las reducciones guaraníticas. En este contexo, se desentrañan los intereses políticos y económicos que dividieron a la sociedad asunceña en dos grupos antagónicos, que se enfrentaron en un conflicto social general en defensa de los privilegios adquiridos. Esta controversia que tuvo imposrtantes repercusiones políticas nos permite adentrarnos en el problema de la mano de obra indígena y en la alianza jesuita-guaraní para comprender el impacto de la conquista espiritual en la consolidación del nuevo orden social.  Este artigo tem como objetivo investigar o impacto da criação do sistema de redução jesuítica nas relações interétnicas hispano-guaranis na região do Paraguai durante a primeira metade do século XVII. Para tanto, analisa o conflito religioso que opôs o bispo franciscano Bernardino de Cárdenas aos padres jesuítas da Companhia pelo controle das reduções guaranis durante a década de 1640. Nesse contexto, desvenda os interesses políticos e econômicos que dividiram a sociedade assuntina em dois grupos antagônicos, que se chocaram em um conflito social generalizado em defesa de seus privilégios adquiridos.
