Materials and Methods: aorta rings were placed in a 20 mL chamber with modified Krebs-Henseleit solution (KHM) balancing out for 60 min. Isometric vessel tension was measured with a force displacement transducer connected to a polygraph. Subsequently, the rings were contracted with 70 mM K+, repeating the procedure until a stable maximal contraction was achieved; post-stabilization for 10 min, vessels were contracted with phenylephrine (0.1µM) and, upon reaching a stable maximum contraction, PLc was added at increasing concentrations (7.14 x 10-6; 7.14 x 10-5; 7.14 x 10-4; 7.14 x 10-3; 7.14 x 10-2; 7.14 x 10-1; and 71.4 µg/mL) so as to create a concentration-response curve. Then, the vessels were again contracted with K+ and finally contracted with for the acetylcholine test (ACh; 1 µM). Results: The concentration of PLc that induced maximal relaxation was 71.4 µg/mL. The PLc concentration producing 50% relaxation of 6.86 µg/mL (n=4) was estimated using a 4-parameter logistic equation (R2 = 0.92). Discussion: PLc has an endothelium-dependent vasodilator effect, the mechanical removal of the endothelium abolishes the relaxant activity of PLc. It reached a magnitude equivalent to the maximal ACh-induced response of about 80% of basal tension. Conclusion: Results support the popular use of Lc. as a hypotensive. Objetivo: Investigar el potencial efecto vasoactivo PLc sobre anillos de aorta aislada de ratas(4,5). Materiales y métodos: Los anillos fueron colocados en una cámara de 20 mL con solución de Krebs-Henseleit modificada, equilibrándose durante 60 min. Medimos la tensión isométrica de los vasos con un transductor de desplazamiento de fuerza conectado a un polígrafo. Posteriormente, los anillos fueron contraídos con K+ 70 mM, repitiendo el procedimiento hasta conseguir una contracción máxima estable; post-estabilización de 10 min., los vasos fueron contraídos con fenilefrina (Phe; 0,1µM) y, al alcanzar una contracción máxima estable, se agregó PLc en concentraciones crecientes (7,14 x 10-6; 7,14 x 10-5; 7,14 x 10-4; 7,14 x 10-3; 7,14 x 10-2; 7,14 x 10-1; y 71,4 µg/mL) construyendo una curva concentración-respuesta. Luego, los vasos fueron contraídos nuevamente con K+ y finalmente con Phe para el ensayo de acetilcolina (ACh; 1 µM). Resultados: La concentración de PLc que indujo la relajación máxima fue 71,4 µg/mL. La que produce un 50% de relajación (EC50) de 6,86 µg/mL (n=4) fue estimada a través de una ecuación logística de 4 parámetros (R2 = 0.92). Discusión: PLc posee efecto vasodilatador dependiente de endotelio, su remoción mecánica anula esta actividad. El efecto alcanzó una magnitud equivalente a la respuesta máxima inducida por ACh, de un 80% respecto a la tensión basal Conclusión: Esto apoyaría el uso popular como hipotensor.
