It is the partial anua letter of 1621 of the reduction of Encarnación de Itapúa written by Fr. Diego de Boroa to the provincial Pedro de Oñate. The transcription is accompanied by a first analysis that seeks to clarify the dating of the transfer of the main settlement of the said reduction from the left bank of the Paraná River to the right bank where it was to become its definitive settlement. In addition, other events reported in the document, such as plagues, famines, and missionary ventures to the Iaña, Iguazú and Guairá, are also described. El presente trabajo está enfocado en el análisis de una fuente jesuitas que permanecía inédita hasta esta transcripción. Se trata de la carta anua parcial de 1621 de la reducción de Encarnación de Itapúa que escribió el P. Diego de Boroa al provincial Pedro de Oñate. Acompaña la transcripción un primer análisis que busca esclarecer la datación del traslado del asiento principal de dicha reducción desde el margen izquierdo del río Paraná hacia el margen derecho donde será su asiento definitivo. Además, se reseñan otros acontecimientos cuya noticia trae el documento como ser, pestes, hambrunas, emprendimientos misionales al Iaña, Iguazú y Guairá.
