The phenotypes adopted by immune cells in their responses are influenced by the "mitochondrial dynamics" (MD): set of characteristics of shape, position and size of mitochondria. These organelles are currently considered to have regulatory functions upon innate and adaptive immunity. The T-lymphocytes intervene in the adaptive response and upon activation they express the CD45RO marker molecule. The endotoxemia produced by endotoxins such as lipopolysaccharide (LPS) in the blood circulation lead to inflammation in multiple organs. T-lymphocytes respond innately to LPS with signaling via the TLR4 receptor, it does not affect proliferation or cytokine secretion but increases adherence and inhibits their migration. The aims of the present work were to observe the morphological characteristics of mitochondria in lymphocytes in cultures of leukocytes exposed to LPS, formylated peptides (fMLP) or ovalbumin (OVA) and mark CD45RO in tests with LPS or OVA.
Autologous leukocyte cultures from healthy heparin anticoagulated human blood samples (n = 10) with ethical consent (HNC, FCM), were stimulated with 25 ng / ml of LPS or 0.25 ng / ml of fMLP or 100 ug / ml of OVA, for 30 minutes. Immunofluorescence technique with anti-CD45RO, DNA staining with DAPI. Paired blood samples provided the controls. Cultured cells were studied with transmission electron microscopy.
Alterations of the mitochondrial morphology of the lymphocytes were observed in the samples with LPS with an increase in the size of the mitochondria and the complexity of the cristae, with electrolucid images (t test for paired samples, p <0.0001). Significant differences were observed in the increased mitochondrial areas in the OVA test (p <0.005), but not in the fMLP assay. Increased positivity was observed for CD45RO in trials with LPS (p <0.005) or OVA (p <0.0001) with respect to paired samples without stimulation.
The increase of the mitochondrial area has been related to early activation of T-cells in the OVA assay, coinciding with CD45RO positivity. The various activators influence the MD and the phenotypes of the immune cells and such MD is important both in the metabolic and immune functions in said cells.

Los fenotipos adoptados por las células inmunitarias en sus respuestas están influenciados por la "dinámica mitocondrial" (DM): conjunto de características de forma, posición y tamaño de las mitocondrias. Estos orgánulos se consideran actualmente con funciones reguladoras de la inmunidad innata y adaptativa. Los linfocitos T intervienen en la respuesta adaptativa y al activarse expresan la molécula marcadora CD45RO. La endotoxemia producida por endotoxinas como el lipopolisacárido (LPS) en la circulación sanguínea conducen a inflamación en múltiples órganos. Los linfocitos T responden innatamente al LPS con señalización vía el receptor TLR4, no afecta la proliferación, ni la secreción de citoquinas pero aumenta la adherencia e inhibe su migración. Los objetivos del presente trabajo fueron observar las características morfológicas de las mitocondrias en linfocitos en cultivos de leucocitos expuestos a LPS, péptidos formilados (fMLP) u ovoalbúmina (OVA) y marcar CD45RO en ensayos con LPS u OVA. Se estimularon cultivos autólogos de leucocitos de muestras de sangre humana sana (n = 10) con consentimiento ético (HNC, FCM), anticoagulados con heparina, con 25 ng/ml de LPS o 0,25 ng/ml de fMLP o 100 ug/ml de OVA, por 30 minutos. Técnica de inmunofluorescencia con anti-CD45RO, tinción de ADN con DAPI. Muestras de sangre apareadas proporcionaron los controles. Se estudiaron las células de los cultivos con microscopía electrónica de transmisión. Se observaron alteraciones de la morfología mitocondrial de los linfocitos en las muestras con LPS con un aumento en el tamaño de las mitocondrias y la complejidad de las crestas con imágenes electrolúcidas (prueba t para muestras apareadas, p <0,0001). Se observaron diferencias significativas de las áreas mitocondriales aumentadas en el ensayo con OVA (p <0,005) no así en el ensayo con fMLP. Se observó positividad incrementada para CD45RO en ensayos con LPS (p<0,005) u OVA (p<0,0001) con respecto a las muestras apareadas sin estimulación. El aumento del área mitocondrial se ha relacionado con activación temprana de células T en el ensayo con OVA, coincidente con positividad de CD45RO. Los diversos activadores influyen en la DM y en los fenotipos de las células inmunes y tal DM tiene importancia tanto en las funciones metabólicas como inmunes en dichas células.
