Therefore, we have calculated the extent of discrimination against [I-aH]Gl-c 6-P by glucose-6-phosphate dehydrogenase as in the experiments with the purified enzymes with the realization that the calculation provides only an indication of this discrimination and not an exact estimate (Table II). The value calculated for the quantity of tritiated substrate remaining unoxidized in the experiment with 164 m/~nloles of TPN + was slightly greater than the amount of Glc-6-P added. The difference is within the limits of experimental error (less than 4 %). These calculations show that in the experiments with the mammary gland glucose-6-phosphate dehydrogenase, the first carbon of the tritiated hexose phosphate was not appreciably oxidized until more than 3o °'o (Table II, 82/27o ) of the ~4C-labeled substrate was utilized. Furthermore, the extent of oxidation of the [I-aHqGlc-6-P was only 1/3 that of tile 14C-!abeled hexose phosphate when 45 % (Table II, 12o/27o ) of tile ('-I of the substrate had been oxidized. We, therefore, concur with the conclusions of KATZ et al. 4,~ who state that ~I-aH~glucose should not be the only all-labeled compound used to estimate the pathways of hydrogen in biological systems. This investigation was supported by a grant from tile National Science Foundation to S. A.
