La calabaza (Cucurbita moschata Duchesne ex-Poiret) fue utilizada para formular un snackfortificado con hierro y Lactobacillus casei ATCC-393. Se determinó que el 55% del hierro resultóbioaccesible a nivel intestinal, y que su presencia y el almacenamiento afectaron tanto laviabilidad del probiótico, >10 7 UFC/g de calabaza durante 14 días a 8°C, como su tolerancia a ladigestión gastrointestinal in vitro (DGI). El producto fortificado obtuvo buena aceptabilidadglobal y el agregado de hierro no fue percibido. Los subproductos del proceso anterior, pulpa y cáscara de calabaza, fueron satisfactoriamenteutilizados como sustrato-soporte de Lactobacillus casei presentando un recuento viable >10 8 UFC/g. La supervivencia del probiótico luego de la estabilización y de la DGI fue >74% durante 28 días de almacenamiento a temperatura ambiente. En general, las propiedades físico-químicasy funcionales del polvo de calabaza (PC) no se vieron afectadas por la presencia del probiótico. El nuevo ingrediente a base de PC conteniendo L. casei fue testeado como suplemento dietarioen dos bebidas comerciales. Se observó un incremento de la viscosidad y un porcentaje desupervivencia >78% del probiótico después de la DGI. Además, se redujo la velocidad deinactivación del probiótico a 63°C respecto del suplemento con células libres. Las bebidas con el PC conteniendo L. casei fueron bien calificadas en cuanto a aroma, textura y aceptabilidadglobal. Por otro lado, el PC conteniendo L. casei fue testeado como ingrediente alimentarioincorporándolo a la formulación de un postre de origen no lácteo. Se obtuvo un postre con unrecuento de probióticos de 10 9 UFC/mL, mostrando además un efecto potenciador del color, lafirmeza y la capacidad de retención de agua, confirmando su potencial aplicación comoingrediente en la industria alimentaria. En conclusión, esta tesis presenta evidencia sobre la potencial utilización de tejido vegetal decalabaza, Cucurbita moschata Duchesne ex-Poiret, para la fortificación con hierro y L. casei ATCC-393, obteniendo un producto con buenas características nutricionales y organolépticas; yla utilización de los subproductos como sustrato y soporte del probiótico para la obtención deun nuevo ingrediente con mayor resistencia a factores de estrés tecnológicos y biológicos,buenas propiedades funcionales, y con alto contenido en fibra dietaria. |

The pumpkin (Cucurbita moschata Duchesne ex Poiret) was used to formulate a ready to eatsnack fortified with iron and Lactobacillus casei ATCC-393. It was determined that 55% of theiron was bioaccessible in the intestine, and that the iron presence and the storage time affectedthe viability of the probiotic, >10 7 CFU/g pumpkin for 14 days at 8 °C, and their tolerance to invitro gastrointestinal digestion (GID). The fortified product presented a good overallacceptability and the addition of iron was not perceived. The byproducts obtained from the previous process, pumpkin pulp and peel were successfullyused as substrate-support for the Lactobacillus casei, showing a viable count >10 8 CFU/g. Theprobiotic survival after the stabilization and the GID was >74% for 28 days of storage at roomtemperature. In general, the physicochemical and functional properties of pumpkin powder (PP)were not affected by the presence of the probiotic. The new ingredient based on PP containing L. casei was tested as a dietary supplement in twocommercial beverages. The viscosity increased and the survival percentage of the probiotic was >78% after GID. Furthermore, the inactivation rate of the probiotic as a supplement was reducedat 63 °C respect to the free cells. The beverages supplemented with PP containing L. casei werewell qualified in terms of flavor, texture and overall acceptability. On the other hand, the PP containing L. casei was tested as a food ingredient in the formulationof a non-dairy dessert. The dessert had a probiotic count of 10 9 CFU/mL, showing besides apotentiating effect of color, firmness and water retention capacity, confirming its potentialapplication as an ingredient in the food industry. To conclude, this thesis presents evidence of the potential use of a vegetal tissue, Cucurbitamoschata Duchesne ex-Poiret pumpkin, for the fortification with iron and L. casei ATCC-393,obtaining a final product with good nutritional and organoleptic characteristics; and the use ofthe byproducts as substrate and support of the probiotic to obtain a new ingredient with higherresistance to technological and biological stress factors, good functional properties, and rich indietary fiber.
