El hígado posee una notablecapacidad regenerativa; frente a un daño extenso o reiterado la regeneración se producepor la activación de las células troncales hepáticas. Estas células se originan durante laembriogénesis hepática y se mantienen latentes durante toda la vida adulta delindividuo. La historia del estudio de las células troncales hepáticas ha estado marcadapor diversas controversias en torno a su existencia, su origen e identificación. Objetivo:aislar, identificar y caracterizar células troncales hepáticas y evaluar la potencialidad dela utilización de éstas como herramienta en el tratamiento de enfermedadesdegenerativas del hígado. Resultados: las células troncales/progenitoras hepáticas (LSPC) de rata expresaron OC2, OC3 y Thy-1, y fueron capaces de proliferar (dandoorigen a las células tipo ovales) cuando se cultivaron in vitro en presencia del factor decrecimiento de hepatocitos (HGF). La expresión de EpCAM de las LSPC de ratón seutilizó como criterio de selección para su aislamiento. La subpoblación EpCAM (+)presentó (por PCR en tiempo real) una expresión significativamente superior decitoqueratina 19, Claudina3, alfa-fetoproteína y Dlk-1. La subpoblación EpCAM (+)mostró una distribución espacial característica de secuencias repetitivaspericentroméricas y un patrón homogéneo de acetilación (K14, K23) y metilación (K9)de la histona H3. Las células EpCAM (+) fueron capaces de anidar en el bazo y migrarhacia el hígado luego del trasplante intraesplénico en ratones. Conclusión: las células EpCAM (+) presentan un patrón de expresión de genes característico y unaorganización nuclear particular que se correlaciona con el carácter troncal que se lesatribuye a las LSCP. En este trabajo se presenta un método sencillo y eficiente paraaislar LSPC de ratón.

Stem cells are defined by their capacity of self-renewal and by the potential to give riseto differentiated cell types. The liver has a remarkable regenerative capacity; when thedamage is extended or repeated the regeneration is carried out by the activation of theliver stem cells. These cells are originated during the hepatic embryogenesis and theystay latent during the whole life of the individual. The history of the study of liver stemcells was characterized by controversies around their existence, their origin andidentification. Objective: to isolate, identify and characterize liver stem cells and toevaluate their potential use as tools for the treatment of degenerative disorders of theliver. Results: the liver stem/progenitor cells (LSPC) from rat expressed OC2, OC3 and Thy-1, and they were capable of proliferate (giving rise to oval-like cells) when theywere cultured in vitro in presence of hepatocyte grow factor (HGF). The expression of EpCAM by the LSPC was used as selection criteria for the isolation. The subpopulation EpCAM (+) showed (by Real-time PCR) a significantly superior expressionof citoqueratin 19, claudin3, alpha-fetoprotein and Dlk-1. The sub-population of EpCAM (+) cells presented a characteristic spatial distribution of the repetitivesequences peri-centromeric and a homogeneous pattern of acetylation (K14, K23) andmethylation (K9) of the histone H3. The EpCAM (+) cells were able to home in thespleen and migrate to the liver after the intra-splenic transplant in mice. Conclusion: EpCAM (+) cells exhibit a characteristic pattern of gene expression and a distinctivenuclear organization that correlate with the stemness attributed to LSPC.
