Peripheral blood mononuclear cells were obtained and expanded using Nef and Gag peptide pools. Post-expansion, CD8TC specificity, functionality, memory phenotype differentiation, PD-1 expression, as well as antiviral activity were assessed. Even though, post-cART HIV specific immune response results deeply diminished, it could be selectively expanded, demonstrating a polifunctional and effector memory/effector profiles, as well as strong antiviral activity. Differences in cART initiation timing affected mainly memory phenotype differentiation, possibly reflecting the impact of antigenic stimulation on the immune system. In both groups, characteristics of the immune response observed before cART could be proposed as biomarkers of the functionality of CD8TC that persist post-cART. El objetivo de esta tesis fue investigar la funcionalidad y el fenotipo LTCD8 HIV-específicos expandidos in vitro, obtenidos de individuos HIV+ bajo tratamiento antirretroviral combinado (cART). Según el momento de inicio del cART, se reclutaron dos grupos, grupo de inicio del tratamiento temprano (TTe) o tardío (TTa). Se obtuvieron células mononucleares de sangre periférica y fueron expandidas con pooles de péptidos de Nef y Gag. Post-expansión se evaluó sobre los LTCD8 la especificidad, funcionalidad, diferenciación del fenotipo de memoria, expresión de PD-1, y la actividad antiviral. A pesar de que la respuesta se encuentra disminuida post-cART, ésta pudo ser específicamente expandida in vitro, demostrando un perfil polifuncional y de memoria efectora, y elevada capacidad de mediar actividad antiviral. El momento de inicio del tratamiento impactó en la diferenciacion del fenotipo de memoria, posiblemente reflejando diferencias en la duración de la estimulación antigénica sobre el sistema inmune. En ambos grupos, se encontraron características de la respuesta inmune antes del cART que podrían actuar como biomarcadores de las células que persisten post-cART.
