Sumario:The present doctoral thesis is about the evaluation of different routes of administration\nof chemotherapeutic agents with possible translation to the treatment of retinoblastoma.\nDifferent routes of administration of topotecan and melphalan were evaluated including\ntopical, intravitreal, intravenous (IV) and superselective intraarterial ophthalmic infusion\n(SIAO).\nThe toxicity and pharmacokinetics of topotecan and melphalan were evaluated after\nSIAO infusion in patients with intraocular retinoblastoma. In addition, the bioavailability of\ntopotecan in the optic nerve of pigs was evaluated after SIAO administration and\ncompared to that obtained after IV infusion as a possible translation to the treatment of\npatients with disseminated disease. A topotecan ointment was also developed and\ncharacterized in vivo with potential use in cases of anterior chamber dissemination. Finally,\na sustained release topotecan loaded hydrogel was developed and characterized as a\nproof of concept for continuous exposure to the drug in the vitreous humor in vitro and in\nvivo. The results obtained in this thesis support the use of the studied routes in the\ntreatment of the discussed ocular tumor.
