| Sumario: | There is evidence that human sperm cryopreservation altered its natural\nphysiological parameters, however it may induce damage to nuclear integrity. This\nwould result in a decrease of viability and fertilizing ability of sperm samples that\nwould be subsequently used in reproduction techniques.\nThis study aims to assess whether damage occurs in sperm nucleus of men\nthat don´t consult for infertility and their sperm have being exposed to\ncryopreservation. 44 samples from healthy men under 40 years old that are\ncandidates for a sperm donation program of a sperm bank at the Argentine Republic\nwere used. A basic sperm analysis was performed for each sample and chromatin\npackaging was assessed with CMA3 and Aniline blue. Samples are subsequently\nfrozen at slow rates in liquid nitrogen and got thawed for further analysis. Nuclear\nintegrity was assessed using TUNEL test, showing evidence of DNA fragmentation at\nthree phases: before freezing, after thawing and after thawing and selected by a\nsperm separation gradient. Data shows normal distribution and was performed using\nSPSS software.\nResults from the current investigation show that cryopreservation induce a\ndeleterious and significant effect at nuclear sperm integrity of healthy and young\nmen, we emphasize on the importance of a sperm selection method after\nfreezing/thawing techniques to reduce the percentage of sperm with structural and\nmolecular injury rates. |
