| Sumario: | Hipertensión is known for being a desease of high prevalence World-wide, commonly related to death related to cardiovascular events. DOCA-Salt rat is an animal modelo f hipertensión, characterized by an increase in plasmatic and central endothelins levels (ETs), and in the sympathetic activity. Recently we proved that ETs could acta neuromodulating catecholaminergic neurons in the ofactory bulbs (OB), anterior hypothalamus (AH), acknowledged as a sympathetic inhibitor area, and posterior hypotahalmus (PH), a sympathoexcitatory center. Hence, we hypothesized that central ETs are envolved in the hipertensión of DOCA-Salt rats, affecting catecholaminergic activity. Initially we proved that ETs receptors ET-A and ET-B are expressed in the regions of interest, and that both colocalize with tyrosine hydroxylase (TH) mostly in the OB and PH of hypertensive rats. In adition, we injected centrally an ET-A antagonist (BQ-610) ora n ET-B agonist (IRL-1620). An tour later rats were sacrificad. Both durgs decreased blood pressure heavily, mainly in hypertensive rats. On the other hand, both BQ-610 and IRL-1620 diminished TH activity, the rate limiting enzyme in catecholamine síntesis, in OBs and PH of hypertensive rats. According results were found in TH phosphorylation, protein expresión and mRNA. Thus, we showed the importante of central endogenous endothelinergic system on maintaining blood pressure levels on DOCA-Salt rats. |
