Sumario:Peritubular myoid cells (PM cells) are found in the wall of seminiferous tubules (ST) and participate\nin ST contraction. These cells have an atypical cytoskeleton organized in two independent layers: the\ninner layer and the outer layer with myofilaments perpendicular and parallel to the ST axis,\nrespectively. This model differs from the orthogonal mesh of filaments previously proposed for PM\ncells. Therefore, in addition to the organization of cytoskeleton, the distribution of other structures\nand proteins involved in the contraction were evaluated in adult rats. The effects of endothelin-1 on\nST, especially on PM cells, were analyzed. In postnatal testicular development, the cytoskeleton\nmaduration and the start of the contractile response were assessed. By different microscopes, it was\nobserved that PM cells despite being flat cells, have a very organized machinery, with structures such\nas endothelin receptors and caveolae orientated along two myofilaments layers that coexist in the\nsame cell, allowing the cell to contract in two directions. In the development, the myofilaments\norganization changes depending on progress of spermatogenesis. The two layers do not appear\nsimultaneously. Even with one of the myofilaments layers, the PM cells are able to contract\nseminiferous cords in response to endothelin-1.
