This technique is a modification of the customary technique, consisting in preparing the tissular stratum or bed from which the sample is to be t:aken. Then, toluidine blue stain is used to outline the most representati e areas of atipical · cells and discariosis.The results of the present work showed the efficacy of this method, specially conceming leucoplakia and verrucous lesions. Atipical cells and discariosis were found in 18,82% of cases, while the biopsy showed the presence of lesion in 21,17 o/o of cases. The cellular flaps from carcinomas (Papanicolau's Grade IV and V) showed evidence of an intercellular substance characterized by being P AS positive, alcian blue pH 2,5 positive, metachromatic to toluidine blue pH 3,8 and 7 and aldehido-fucsine po~itive. This intercellular substance conceals the cells of the lesion. Se estudiaron 140 lesiones bucales sospechosas de malignidad, empleando la citología "por desgarro". Esta técnica, que es una modificación de la técnicaclásica o convencional, está basada en la preparación del lecho para la toma del material; luego de este primer paso el empleo del azul de toluidina puede indicar o marcar áreas de la lesión representativas i!e atipías celulares y discariosis, de donde se toma el material. Los resultados que se obtuvieron ponen en evidencia la eficacia del método, principalmente en lesiones leucoplásicas y verrugosas, con 18,82% de casos en los que se han detectado células atípicas y discariosis, mientras la biopsia las demostró en 21,17% de los casos. Los colgajos celulares obtenidos de carcinomas (grado IV y V de Papanicolaou) pusieron de manifiesto la presencia de una sustancia intercelular que enmascara las células y que se caracteriza histoquímicamente por ser PAS positiva, alcian blue a pH 2,5 positiva, metacromática al azul de toluilina a pH 3,8 y 7 y aldehído fucsina positiva.
