description Lipoic Acid (LA) is an antioxidant that reaches fetal circulation after maternal oral intake. Furthermore, the fetal period is characterized by strong changes in the expression of genes that, if altered they can influence the organism fenotype in the postnatal life. The aim of the present work was to assess the effect of maternal intake of AL on gene expression related to the anti-oxidant activity and regulation of fatty acids in the liver of ovariectomized rat offspring. Pregnant Wistar rats were fed ad-libitum with standard diet (food) + LA (0.4% wt/wt) from day 14 of gestation to day 20 of lactation (L, n=4) or food (C, n=4) as control. Litter size was reduced to 3 females after birth. At 8 months of age, female offsprings were randomly assigned to be ovariectomized (OV) or to receive a sham surgery (SH); resulting in 4 groups: LOV, LSH, COV and CSH (n=5/group). Animals were euthanized after two months for samples’ collection. Expressions of hepatic genes involved in sterol synthesis (Srebf1), oxidation of fatty acids (Cpt1a) and antioxidant enzymes (Cat and Sod1) were measured through qRT-PCR. Results were analyzed by ANOVA and significance was set at p<0.05. Compared to CSH group, the expression of Srebt1 was higher in COV group; Cpt1a and Sod1 were higher in LSH group and Cat was higher in LOV and LSH group. Expression of liver genes involved in sterol synthesis, oxidation of fatty acids and antioxidant enzymes were modified by the maternal intake of LA, possibly due to the effect of fetal programming. Therefore, the maternal intake of LA improves the oxidative state of the offspring liver, even when it can be affected by the deficiency of estrogen induced by ovariectomy

Las propiedades antiobesogénicas de AL vienen dadas por una modificación de los genes relacionados con el metabolismo y el gasto energético. Por otro lado, el período fetal se caracteriza por fuertes cambios en la expresión génica que, si se alteran, pueden influir en el fenotipo del organismo en la vida postnatal. El objetivo del presente trabajo fue evaluar el efecto del consumo materno de AL sobre la expresión de genes relacionados con la actividad anti-oxidante y regulación de ácidos grasos en el hígado de las crías ovariectomizadas. Ratas Wistar preñadas se alimentaron ad-libitum con alimentos molidos + AL (0.4% p/p) desde el día 14 de gestación hasta el día 21 de lactancia (L) o alimentos molidos (C) como control. El tamaño de la camada se redujo a 3 hembras después del nacimiento. A los 8 meses de edad, las crías hembras fueron asignadas aleatoriamente para ser ovariectomizadas (OV, n=10) o para recibir una cirugía simulada (SH, n=10) resultando en 4 grupos: LOV, LSH, COV y CSH (n=5 por grupo). Después de 2 meses, los  animales fueron eutanasiados para la colección de las muestras. La expresión de genes hepáticos involucrados en la síntesis de esterol (Srebf1), oxidación de ácidos grasos (Cpt1a) y enzimas antioxidantes (Cat y Sod1) fueron medidos a través de qRT-PCR. Los resultados fueron analizados por ANOVA, y se consideraron estadísticamente significativos a un p<0.05. Comparado con el grupo CSH, la expresión de Srebt1 fue mayor en el grupo COV; Cpt1a y Sod1 fue mayor en el grupo LSH y Cat fue más alta en los grupos LOV y LSH. La expresión de genes hepáticos involucrados en la síntesis de esterol, oxidación de ácidos grasos y enzimas antioxidantes fue modificada por la ingesta materna de AL, debido posiblemente al efecto de la programación fetal. Por ende, el consumo materno de este mejora el estado oxidativo del hígado de la crías, aun cuando este puede ser afectado por la deficiencia de estrógenos inducida por ovariectomía Universidad Nacional Cba.
